Service d'imagerie cellulaire in vitro et ex vivo

L’immunofluorescence observée en microscopie confocale permet de localiser les éléments cellulaires tels que protéines, chaînes lipidiques, ions, et acides nucléiques à l’intérieur de la cellule. Cependant, ce marquage doit être effectué sur des spécimens qui ont été préalablement soumis à une fixation. Alors, les évènements observés sont le reflet d’un seul moment dans le temps sans aucun dynamisme temporel. De plus, le fait que les différents échantillons à comparer soient traités individuellement rend l’analyse quantitative difficile et elle doit être supportée par des expériences biochimiques, électrophysiologiques et de biologie moléculaire.

L’observation confocale en fluorescence sur cellules vivantes permet de visualiser et quantifier avec précision des phénomènes dynamiques dans le temps. Cette visualisation doit s’effectuer sans affecter la viabilité cellulaire, ce qui est primordial dans le but d’obtenir des résultats exacts. Les manipulations doivent donc s’effectuer sous conditions physiologiques similaires à celles présentes in vivo.

Calcium intracellulaire, membranes, organelles, radicaux libres, FRAP, photo-manipulations, protéines fluorescentes, analyses de migration, reconstructions en 4-dimensions, interactions cellules-cellules sont des exemples d’investigations effectuées sur des cellules vivantes avec les microscopes confocaux du Centre de Recherche de l’Institut de Cardiologie de Montréal.

Objectifs
  1. Acquisition , traitements et analyse d’images confocale en fluorescence pour les équipes de recherche œuvrant au Centre de Recherche de l’Institut de Cardiologie de Montréal et de leur collaborateurs.
  2. Développement de techniques d’imagerie cellulaire spécifiques pour chaque projet de recherche
  3. Entraînement des chercheurs/étudiants/membres du personnel de recherche aux techniques de marquage en fluorescence, à l’utilisation des microscopes confocaux ainsi qu’aux logiciels d’analyse/reconstruction 3D.

  1. Acquisition , traitements et analyse d’images confocale en fluorescence pour les équipes de recherche œuvrant au Centre de Recherche de l’Institut de Cardiologie de Montréal et de leur collaborateurs.
  2. Développement de techniques d’imagerie cellulaire spécifiques pour chaque projet de recherche
